We offer fragment analysis of various types of your prepared fluorescently labelled DNA fragments (microsatellite analysis, RFLP - Restriction Fragment Length Polymorphism, AFLP - Amplified Fragment Length Polymorphism, MLPA - Multiplex Ligation-dependent Probe Amplification and other applications).
Fragments labeled with one or more dyes are analyzed on a genetic analyzer with sixteen capillaries (3130xl Genetic Analyzer, Applied Biosystems), located in Viničná 7.
The instrument has spectral calibrations set using the following standards:
- DS-30 (6FAM™, HEX, NED™, ROX™) for the red standard ROX400 and ROX500.
- DS-33 (6FAM™, VIC®, NED™, PET®, LIZ®) for the orange standard LIZ500,LIZ600,LIZ1000.
- DS-02 (dR110, dR6G, dTAMRA™, dROX™, LIZ®) for orange standard LIZ120 for SNaPshots
Sample preparation:
We accept samples already prepared for analysis and denatured.
Mix the samples (0,5 μl) with formamide (9 -9,5 μl) and add your own standard (0,25 - 0,5 μl). Follow the techniques established in your laboratory. Observe a minimum volume of 10 μl in the plates or 10,5 μl in the stripes. We do not supply internal standards (ROX and LIZ).
Deliver samples in strips or in an optical plate. The assay runs on 16-capillary platforme, so try to keep the number of samples to a multiple of 16 if possible. Optical plates with special caps and formamide can be obtained in advance from our laboratory.
Samples should be left in Viničná 7 in the corridor in the 2nd basement in a small fridge in the freezer shelf. Samples are processed virtually every day.
Fill in Sample sheet for Fragment Analysis and send it to seqlab@natur.cuni.cz or to hrda@natur.cuni.cz. The printed sample sheet do not need to be sent with the samples. The sample sheet is partially self-filling - if not, download it again.
Name the sample sheet, the subject of the E-mail, and the sample bag with the abbreviation and date in the form FFSDDMMRR-fr. E.g.: 6. 9. 2024 - Ford Harrison, will be marked HAF060924-fr.
Label individual tubes with the abbreviation and serial number from the sample sheet. We recommend to use strips or plates for larger number of samples (you can request them in advance). Number logically.
Please note: 1 sample = 1 tube/well = 1 line in the sample sheet = 1 analysis.
