Genolab is a facility shared among the Dpt. of Zoology, Dpt. of Ecology, and Dpt. of Botany. The main purpose of the laboratory is to work with clean next-generation sequencing methods. Currently, the protocols running are


• RNA extraction
• NGS library preparation (for RNA-seq, whole genome seq, RAD-seq)
• Single-cell library preparation


It is possible to introduce new protocols to the laboratory upon agreement. No PCR products or live/fresh tissues are allowed. (Unfortunately, this also includes metabarcoding approaches based on the PCR steps). Currently, no protocols involving RNAse steps are allowed either.

• Flow box for RNA extraction
• NGS library preparation box
• Tissue homogenizer (SpeedMill PLUS)
• Centrifuge (Eppendorf)
• Bioanalyzer (Agilent 2100) – for assessing RNA and DNA quality, integrity, and
fragment size distribution
• Vacuum concentrator (SpeedVac DNA 130) – for concentrating nucleic acid
and protein samples without high heat
• Extraction robot (PurePrep 32) – for nucleic acid extraction, magnetic bead-
based
• Extraction robot (CyBio FeliX) – for automated nucleic acid extraction and
other sample preparation protocols and pipetting tasks
• Pipetting robot (LABOT) – for precise pipetting tasks
• Thermal cyclers (Eppendorf, BIOER)
• Chromium X (10x Genomics) – for library preparation for single-cell and spatial
omics applications
• Thermoblock
• Heat sealer for plates (Eppendorf)
• Freezer -80 °C (Upright Thorbi Primus)


Access to the lab:
New users always need to contact Zuzana Konvičková and register with her before working in the laboratory. Please, make sure that everyone from your group (students, postdocs, and stad members) who works in the laboratory is registered. The lab has chip access. You can have it installed on your ISIC / ITIC card if you fill in a request form for room S004 at Viničná 7 here: http://web.natur.cuni.cz/www/intranet/formulare/form_cip.php.
You need to generate it in PDF format, print it, and get it signed by Zuzana Konvičková (or Zuzana Musilová) and your department head. Once your document has all signatures, deliver it to CIT (Information Technology Division). You can use the faculty post to send it there.
